The influence of orientin, gentiopikroside and oleanolic acid isolated from Gentiana algida Pall on the state of immune system in the experiment

The purpose of the study is to determine the immune modulating activity of orientin, gentiopikroside and oleanolic acid isolated from the aerial part of Gentiana algida Pall. on the cellular and humoral immune response at the azathioprine immune suppression. Methods. Experiments were carried out on CBA male mice (n=100, 18-20 g). Immune deficiency was modeled by the intragastrical introduction of azathioprine in the dose 50 mg/kg once a day for 5 days. The experimantal groups of animals received orientin, gentiopikroside and oleanolic acid in doses 15, 12 и 4 mg/kg respectively once a day intragastrically for 14 days against the background of azathioprine immunosupression. The intact group received the purified water according to the analogous scheme. The effect of the tested compounds on state of the cellular immunity was evaluated in the reaction of delayed-type hypersensitivity (DTH) according to the standard method of local DTH. Condition of the humoral immunity was estimated by the number of antibody-forming cells determined by the method of local hemolysis by A.J. Cunningham (1965). Statistical analysis was performed using standard methods of variation statistics using parametric Student’s t-test. Results. It was established that the biologically active substances in the following doses: orientin - 15 mg/kg, gentiopikroside - 12 mg/kg, oleanolic acid - 4 mg/kg reduced suppressive action of azathioprine on cell-mediated immune response and antibody response, the index of the delayed type hypersensitivity reaction was 1.3 - 1.7 times increased and the absolute and relative numbers of antibody-forming cells was 1.6 - 1.8 times increased as compared with the control group. The results of the studies have shown that orientin and oleanolic acid render most pronounced immunomodulating effect. Conclusion. The results obtained allow to conclude that the orientin, gentiopikroside and oleanolic acid isolated from the aerial part of Gentiana algida has the marked immune modulating activity that substantiates the expediency of their further study in order to create new effective immune modulators.Downloads
